First a standard curve was made using a stock solution containing 100 micrograms/milliter of acorbic acid. This was diluted with buffer and diluted with 0.1 milliliters of DCPIP and 0.1 mL of solution in a cuvette.
The transmittance was read on a spectrophotmeter at 520 nm.
Samples were diluted in duplicate at 1:1, 1:5, and 1:10 ratios.
1 mL of sample and 0.1 mL of DCPIP was added to the cuvette and the absorbance was read.
This was repeated with a second sample.
Note: calculations were completed using an average of the transmittance of the 1:5 dilutions due to variance in data.
A spectrophotometer works by passing a single wavelength of light through the sample and reading the transmittance.
